A novel cost-efficient alternative to protein A chromatography
Protein A chromatography remains the industry standard for mAbs purification but comes with high cost and challenges with integration in continuous processes. Precipitation combined with VMF offers a robust, efficient, alternative down stream processing, with potential to significantly reduce costs.
Higher concentration – enhanced process performance
Using hIgG as a model media, VMF demonstrated stable high permeate fluxes with minimal fouling. The system maintained stable TMP up to 77.4 L/m²/h during flux stepping experiments, achieving a 90% conversion and thus a 10× concentration factor. Across a range of feed flow rates, VMF consistently enabled high throughput while preserving membrane performance and cleanability (regaining >90% of initial membrane permeability), highlighting its suitability and robustness for continuous bioprocessing applications.
Continuous mAbs purification made easy
VMF demonstrated a highly stable process over 24 hours of operation at a constant permeate flux of 72.9 L/m²/h (85% conversion, 6.67× concentration factor) with no loss of precipitated hIgG and a remarkable low TMP increase during the entire process.
Analyzing the particle size distribution of the redissolved hIgG reveals that using Vibro® Membrane Filtration in single-pass continuous processing preserves the product quality, with no observed aggregate formation and providing a purity profile matching the original feed
